Imzokitug + pumitamig is a combination immunotherapy being developed by Bristol Myers Squibb for the treatment of first-line (1L) non-small cell lung cancer (NSCLC). Imzokitug (BMS-986322) is a recombinant interleukin-15 (IL-15) agonist, specifically a complex of IL-15 and the IL-15 receptor alpha (IL-15Rα) subunit, designed to stimulate the proliferation and activation of natural killer (NK) cells and CD8+ T cells. Pumitamig (BMS-986353) is a monoclonal antibody that targets T-cell immunoreceptor with Ig and ITIM domains (TIGIT), an inhibitory immune checkpoint receptor expressed on T cells and NK cells. By combining an IL-15 agonist with a TIGIT inhibitor, the therapy aims to synergistically enhance the anti-tumor immune response by simultaneously providing a stimulatory signal to effector cells and blocking an inhibitory pathway that often limits their activity in the tumor microenvironment.
