Overview

IN10018 + albumin-bound paclitaxel + gemcitabine is a **combination regimen** under investigation primarily for the treatment of advanced pancreatic cancer. IN10018 is a small molecule focal adhesion kinase (FAK) inhibitor, which targets FAK to disrupt survival and motility signaling in tumor cells. Albumin-bound paclitaxel (nab-paclitaxel, Abraxane) is a nanoparticle formulation of paclitaxel that blocks cell division by stabilizing microtubules and is formulated for improved delivery and reduced toxicity. Gemcitabine is a nucleoside analog that inhibits DNA synthesis, leading to cell death. The regimen is designed to leverage potentially synergistic antitumor activity by combining these mechanisms and partially non-overlapping toxicities. IN10018 is being developed and trialed alongside standard of care chemotherapy (nab-paclitaxel and gemcitabine) in advanced pancreatic ductal adenocarcinoma[1][2][3][4][5][6][8].
