IN10018 + docetaxel is a combination investigational regimen comprising IN10018 (also known as BI 853520 or ifebemtinib), a selective ATP-competitive small molecule inhibitor of focal adhesion kinase (FAK), and docetaxel, a microtubule-stabilizing chemotherapeutic agent. IN10018 inhibits FAK autophosphorylation and interferes with key pathways involved in cancer cell survival, proliferation, migration, and tumor microenvironment modulation, especially impacting the FAK/AKT/mTOR signaling pathway. Docetaxel disrupts microtubule function, leading to mitotic arrest and apoptosis of rapidly dividing cancer cells. The combination is under early clinical investigation for solid tumors, including locally advanced or metastatic gastric or gastroesophageal junction (GEJ) adenocarcinoma. Preclinical and clinical data suggest IN10018 may overcome resistance mechanisms and enhance chemotherapy and immunotherapy efficacy when combined with traditional agents like docetaxel[1][2][4].
