IN10018 + nab-paclitaxel + tislelizumab is an investigational combination regimen comprising three agents: IN10018 (code name BGB-283), a potent and selective small molecule inhibitor of the protein focal adhesion kinase (FAK); nab-paclitaxel (brand name Abraxane), an albumin-bound formulation of the chemotherapeutic agent paclitaxel, which disrupts microtubule function to inhibit cell division; and tislelizumab (brand names Tevimbra and Baizean, code name BGB-A317), a humanized monoclonal antibody against programmed cell death-1 (PD-1) receptor, designed to block immune checkpoint pathways and restore T cell-mediated anti-tumor immunity[1][2][3][4]. The combination is used in oncology clinical trials and primarily investigated for indications such as unresectable or metastatic esophageal squamous cell carcinoma (ESCC) and HER2-negative gastric or gastroesophageal junction adenocarcinoma (GEJ)[1][2][3]. This regimen exploits FAK pathway inhibition to reduce tumor cell survival and invasiveness, chemotherapeutic cytotoxicity by paclitaxel, and immune system activation by checkpoint blockade via PD-1 inhibition.
