Overview

A four-drug combination therapy consisting of **IN10018** (a selective FAK inhibitor), **tislelizumab** (a humanized anti–PD-1 IgG4 monoclonal antibody), **carboplatin** (a platinum-based DNA crosslinker), and **etoposide** (a topoisomerase II inhibitor). This regimen is currently under phase Ib/II clinical investigation as a first-line treatment in adults with **extensive-stage small cell lung cancer (ES-SCLC)**. The combination leverages multiple mechanisms: immune checkpoint inhibition (PD-1 blockade by tislelizumab), disruption of tumor stroma and enhancement of immune infiltration (by IN10018), and direct cytotoxicity against tumor cells (by carboplatin and etoposide). The combination is designed to maximize anti-tumor efficacy through synergy between immunotherapy, targeted therapy, and cytotoxic chemotherapy[1][4][5][7].
