Inavolisib + eribulin is a combination therapy being investigated for the treatment of advanced triple-negative breast cancer (TNBC) harboring PIK3CA mutations. Inavolisib (Itovebi) is a potent, selective, and orally bioavailable PI3Kα inhibitor that specifically targets the p110α isoform and induces the degradation of mutant p110α. Eribulin (Halaven) is a synthetic analogue of halichondrin B that acts as a microtubule dynamics inhibitor, specifically binding to the plus ends of microtubules to inhibit growth. This combination aims to provide synergistic antitumor activity by simultaneously inhibiting the PI3K signaling pathway, which is frequently mutated in TNBC, and disrupting mitotic progression. The combination is currently being evaluated in a Phase II clinical trial (NCT06344104) sponsored by investigator Hu Hai.
