Inavolisib + letrozole is a combination therapy consisting of inavolisib (Itovebi), a potent and selective PI3Kα inhibitor that promotes the degradation of mutated p110α, and letrozole, an aromatase inhibitor. This combination is being investigated for the treatment of PIK3CA-mutated, hormone receptor (HR)-positive, HER2-negative locally advanced or metastatic breast cancer. The combination works by targeting both the PI3K pathway (inavolisib) and estrogen production (letrozole) to inhibit cancer cell growth and proliferation.
