Overview

Inavolisib + paclitaxel is an investigational combination therapy consisting of inavolisib, a highly potent and selective oral PI3K inhibitor that targets and degrades mutated PI3Kα (p110α), and paclitaxel, a well-established microtubule-stabilizing chemotherapeutic agent. This combination is being studied primarily for the treatment of locally advanced or metastatic solid tumors—including PIK3CA-mutated HER2-positive breast cancer—where it aims to improve anti-tumor activity by simultaneously inhibiting the PI3K pathway (a key driver of tumor growth and resistance) and disrupting cell division. Early-phase clinical trials have shown that this regimen is generally well tolerated with no new safety signals identified[1][2][5]. Inavolisib was developed by Genentech/Roche.
