**Inavolisib + palbociclib** is a fixed combination of two oral small molecule drugs co-administered with fulvestrant for the treatment of patients with PIK3CA-mutated, hormone receptor-positive, HER2-negative, locally advanced or metastatic breast cancer. Inavolisib is a highly selective PI3Kα inhibitor, while palbociclib is a selective inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6). The combination, together with fulvestrant, acts via dual blockade of the PI3K/AKT/mTOR and CDK4/6 cell cycle regulatory pathways, targeting two key oncogenic drivers in endocrine-resistant breast cancer. It is indicated primarily in patients with disease progression or recurrence during or after endocrine therapy[1][2][3][4][5][7].
