This is a combination regimen comprising **INBRX-106**, **pembrolizumab**, **pemetrexed**, and **cisplatin**, used or studied in the treatment of advanced or metastatic non-squamous non-small-cell lung cancer (NSCLC), as well as potentially other solid tumors. INBRX-106 is a hexavalent OX40 agonist antibody aiming to stimulate the immune system by activating T cells via OX40 receptor agonism, thereby enhancing anti-tumor immune responses. Pembrolizumab is a monoclonal antibody targeting PD-1 (programmed cell death protein 1), functioning as a checkpoint inhibitor that promotes immune-mediated tumor cell death. Pemetrexed is an antifolate chemotherapy agent that disrupts folate-dependent metabolic processes essential for cell replication, and cisplatin is a platinum-containing chemotherapeutic that causes DNA crosslinking and apoptosis in dividing cells. The combination is designed to synergistically enhance anti-tumor efficacy through immune activation (INBRX-106 and pembrolizumab) and direct cytotoxic effects (pemetrexed and cisplatin), forming a quadruple regimen intended for oncology indications where immunotherapy benefits from additional tumor debulking and immunostimulatory chemotherapy.
