Overview

INBRX-109 + carboplatin is an experimental drug combination comprising **INBRX-109**, a third-generation, recombinant, humanized, tetravalent monoclonal antibody that acts as an agonist of death receptor 5 (DR5, also known as the TNF-related apoptosis-inducing ligand receptor 2, TRAIL-R2), and **carboplatin**, a platinum-based cytotoxic chemotherapy. INBRX-109 selectively activates DR5, leading to multimerization of the receptor, formation of the death-inducing signaling complex (DISC), and subsequent activation of caspase-8 and downstream apoptotic caspases 3 and 7, resulting in programmed cell death in tumor cells. Carboplatin causes DNA crosslinking and cell death, providing a complementary cytotoxic mechanism. This combination is under clinical investigation for the treatment of advanced or metastatic solid tumors, including sarcomas and other cancers[1][2][3][6][7].
