Pemetrexed is a **small molecule antifolate chemotherapeutic** that inhibits folate-dependent enzymes required for DNA synthesis. The combination is evaluated for synergistic antitumor effects, especially in solid tumors such as **malignant pleural mesothelioma, pancreatic adenocarcinoma**, and sarcomas (notably chondrosarcoma and Ewing sarcoma)[3][4][6].
