Overview
**INCA00186 + retifanlimab** is an experimental combination therapy comprising INCA00186, a humanized monoclonal antibody that acts as a CD73 (5'-nucleotidase) inhibitor, and retifanlimab, a programmed death receptor-1 (PD-1) blocking antibody. INCA00186 inhibits the enzymatic activity of CD73 to reduce adenosine-mediated immune suppression in the tumor microenvironment, thereby promoting immune cell activity. Retifanlimab is a checkpoint inhibitor that blocks PD-1 on T-cells, stimulating the immune system to recognize and attack cancer cells. This combination is under early-phase clinical investigation for advanced or metastatic squamous cell carcinoma of the head and neck (SCCHN) and selected gastrointestinal malignancies, particularly in CD8 T-cell-positive tumors. The developer for both drugs is Incyte. Retifanlimab (as Zynyz) is approved as monotherapy for other indications, but not in this specific combination.
