INCB001158 + epacadostat is a combination of two investigational small molecule drugs developed for the treatment of advanced or metastatic solid tumors. INCB001158 is an arginase inhibitor (CB-1158), and epacadostat (INCB024360) is an indoleamine 2,3-dioxygenase 1 (IDO1) inhibitor. The combination is intended to modulate the tumor microenvironment and enhance antitumor immune responses by simultaneously targeting L-arginine depletion (via arginase) and tryptophan metabolism (via IDO1). Clinical studies have evaluated this combination with and without pembrolizumab in patients with advanced solid tumors. Development was halted after interim trial data in melanoma failed to reach key efficacy endpoints, and no new enrollment is ongoing for new cohorts[1][3][5].
