Overview
INCB054828 + paclitaxel is an investigational combination therapy consisting of pemigatinib (INCB054828), a selective small molecule inhibitor of fibroblast growth factor receptors 1, 2, and 3 (FGFR1/2/3), and paclitaxel, a microtubule-stabilizing chemotherapeutic agent. Pemigatinib inhibits FGFR-mediated signaling pathways involved in cell proliferation and survival in cancers with FGFR genetic alterations. Paclitaxel disrupts microtubule dynamics required for mitosis, leading to cell cycle arrest and apoptosis. The combination is being studied as a second-line treatment for recurrent or progressive gastric cancer patients harboring FGFs/FGFRs genetic mutations[9][1]. Preclinical data support the rationale that combining an FGFR inhibitor with chemotherapy may enhance antitumor efficacy by targeting complementary pathways[3][6].
