INCB177054 + retifanlimab

INCB177054 + retifanlimab is an investigational combination therapy for advanced or metastatic solid tumors. INCB177054 is a potent, orally bioavailable dual inhibitor of diacylglycerol kinase alpha (DGKα) and diacylglycerol kinase zeta (DGKζ). It enhances T-cell signaling and activation, lowers the threshold for T-cell activation to tumor antigens, and increases antitumor immune response. Retifanlimab (retifanlimab-dlwr; brand name Zynyz) is an anti–programmed cell death protein 1 (PD-1) monoclonal antibody checkpoint inhibitor, designed to interrupt the PD-1/PD-L1 pathway and enhance antitumor immunity. In preclinical models, INCB177054 and retifanlimab demonstrate synergistic activity. This combination is being studied primarily in adult patients with advanced or metastatic solid tumors who have progressed following standard therapy[1][2][3][5][6][7][9].
