Overview

INCB7839 + trastuzumab is a combination therapy investigated for the treatment of HER2-positive metastatic breast cancer. INCB7839 (also known as INCB007839) is an oral small molecule inhibitor targeting ADAM10 and ADAM17, two metalloproteinases involved in the shedding of ligands that activate EGFR/HER family receptors. Trastuzumab is a monoclonal antibody directed against the HER2 receptor. The rationale for this combination stems from evidence that inhibition of ADAM10/17 can reduce resistance to trastuzumab by decreasing ligand-induced activation of alternative growth pathways and reducing HER2 cleavage. Clinical studies have shown that this combination was generally well tolerated and demonstrated objective response rates between 40% and 55% in patients with advanced breast cancer[3][5][6]. The development was led by Incyte Corporation.
