A combination therapy of **indatuximab ravtansine** (BT062), **lenalidomide**, and **dexamethasone** under investigation for the treatment of **multiple myeloma**, particularly relapsed or refractory cases. **Indatuximab ravtansine** is an antibody-drug conjugate (ADC) consisting of a chimeric anti-CD138 monoclonal antibody (nBT062) linked to the cytotoxic maytansinoid agent DM4, via a disulfide linker. After binding to CD138 (syndecan-1) on malignant plasma cells, it is internalized and the DM4 component is released intracellularly, binding to tubulin and disrupting microtubules, causing cell cycle arrest and apoptosis. Lenalidomide is an immunomodulatory agent (IMiD) that enhances immune response and has antineoplastic properties, while dexamethasone is a synthetic glucocorticoid with anti-inflammatory and immunosuppressive activities. This combination aims to enhance antitumor activity by hitting multiple targets and mechanisms in multiple myeloma cells. The regimen is being researched in phase 1/2 clinical trials for relapsed or refractory multiple myeloma[1][4][7][10].
