indinavir + nelfinavir

**Indinavir + nelfinavir** is a combination of two HIV-1 protease inhibitors, indinavir and nelfinavir, studied as part of multidrug antiretroviral therapy for HIV infection. Both drugs inhibit the HIV-1 protease enzyme, blocking viral polyprotein cleavage and preventing the formation of mature, infectious virions. Used together, they were investigated for pharmacokinetic advantages based on the premise of mutual drug boosting via CYP3A4-mediated interactions, enabling possible twice-daily dosing intervals. However, results show unpredictable pharmacokinetic interactions, and evidence of antiviral activity is limited to small studies. Both agents have independent toxicity profiles, including nephrolithiasis (indinavir) and diarrhea (nelfinavir).
