Indoleamine 2,3-dioxygenase 1 (IDO1) small interfering RNA (siRNA) is a therapeutic modality designed to silence the expression of the IDO1 enzyme through RNA interference. IDO1 is a key enzyme in the kynurenine pathway that degrades tryptophan, leading to an immunosuppressive environment and contributing to chemoresistance in various malignancies, including ovarian cancer. By specifically targeting and degrading IDO1 mRNA, this siRNA approach reduces IDO1 protein levels, which has been shown to increase reactive oxygen species (ROS) accumulation and enhance p53-dependent apoptosis. This mechanism sensitizes cancer cells to platinum-based chemotherapy, such as cisplatin, and may help overcome resistance in type II ovarian cancer patients.
