INDV-2000 + buprenorphine + naloxone is a combination regimen under clinical investigation for the treatment of **opioid use disorder**. INDV-2000 is a potent and selective oral antagonist for the orexin-1 receptor (OX1R), acting as a non-opioid agent that targets the orexin pathway which plays a key role in the neurobiology of substance use disorders[1][3][5]. Buprenorphine is a partial opioid agonist at mu-opioid receptors, managing withdrawal and cravings while exerting a ceiling effect to reduce risk of overdose[2][4][6]. Naloxone is a pure opioid receptor antagonist (primarily mu-opioid) added to deter intravenous misuse and reverse opioid effects in case of overdose[2][4][6]. In clinical protocols, subjects may be stabilized on buprenorphine/naloxone, then treated with INDV-2000 in combination during the withdrawal/transition phase[5]. The combination is being developed and tested by Indivior specifically to assess safety and efficacy in individuals with opioid use disorder.
