This combination therapy consists of inebilizumab, a humanized monoclonal antibody that targets and depletes CD19+ B cells (including plasmablasts and plasma cells), and dazodalibep (VIB4920), a non-antibody fusion protein that inhibits the CD40 ligand (CD40L). Developed by Amgen, the combination is being evaluated in a Phase 2 clinical trial for the desensitization of highly sensitized patients with end-stage renal disease (ESRD) who are awaiting kidney transplantation. By targeting both B-cell depletion and the CD40/CD40L costimulatory pathway, the therapy aims to reduce levels of donor-specific alloantibodies, thereby increasing the likelihood of a successful organ match and transplant.
