Overview

This is a combination of five monoclonal antibody biologics, each with distinct but related mechanisms targeting immune pathways involved in chronic inflammatory diseases. Infliximab, adalimumab, and golimumab are anti-tumor necrosis factor alpha (TNFα) agents that block the activity of TNFα, a cytokine central to inflammation in autoimmune disorders. Vedolizumab is an integrin blocker that selectively inhibits α4β7 integrin-mediated lymphocyte trafficking to the gut, reducing gastrointestinal inflammation. Ustekinumab targets interleukin 12 and interleukin 23 by binding their shared p40 subunit, thereby modulating immune responses implicated in several inflammatory conditions. These drugs are primarily used individually for moderate-to-severe ulcerative colitis and Crohn’s disease when conventional therapies fail[1][6][9]. There is no evidence or clinical precedent for using all five agents together as a single combination therapy.
