Overview

INGN 201 + doxorubicin + docetaxel is an experimental combination therapy consisting of INGN 201 (Advexin), an adenoviral vector-based p53 gene therapy, plus the chemotherapeutic agents doxorubicin and docetaxel. INGN 201 delivers a wild-type p53 gene via a recombinant adenovirus directly into tumor cells, restoring p53 tumor suppressor activity to induce apoptosis, inhibit cell proliferation, and potentially sensitize tumors to chemotherapy. Doxorubicin is an anthracycline antibiotic that intercalates DNA and inhibits topoisomerase II, resulting in cytotoxic effects, while docetaxel is a taxane that promotes microtubule stabilization, preventing cell division and leading to apoptosis. This combination has been studied primarily for advanced, resectable squamous cell carcinoma of the head and neck, with perioperative INGN 201 therapy followed by chemoradiotherapy including agents such as doxorubicin and docetaxel. The regimen is under investigation for improved local tumor control and survival outcomes[1][2][3].
