Overview

INI-4001 + avelumab is an investigational combination therapy being studied for the treatment of advanced solid tumors. **INI-4001** is a proprietary small molecule agonist of toll-like receptor 7 and 8 (TLR7/8) formulated in a nanoparticle delivery system, developed by Inimmune. It is designed to promote antitumor immunity by activating TLR7/8, leading to induction of cytokines such as IFNα, activation of antigen-presenting cells, and subsequent T cell activation. **Avelumab** is a monoclonal antibody targeting programmed death-ligand 1 (PD-L1) and acts as a checkpoint inhibitor. The combination is being tested to determine if co-activation of innate immunity (via TLR7/8 agonism) and inhibition of the PD-1/PD-L1 axis can increase anti-tumor responses in subjects for whom checkpoint inhibition alone is suboptimal. The combination is currently under investigation in a Phase 1 clinical trial for patients with advanced solid tumors[1][2][3][4].
