Overview

Iniparib + paclitaxel is an investigational combination therapy consisting of iniparib and the chemotherapeutic agent paclitaxel. Iniparib was initially developed as a poly(ADP-ribose) polymerase (PARP) inhibitor but later research indicated it does not act as a PARP1 inhibitor at pharmacological concentrations. Its mechanism is now thought to involve targeting redox metabolism and increasing oxidative stress to induce cancer cell death. Paclitaxel is a well-established microtubule-stabilizing agent that inhibits cell division by promoting tubulin polymerization and preventing microtubule disassembly. The combination has been studied primarily for triple-negative breast cancer (TNBC) and uterine carcinosarcoma, but clinical trials have shown no significant improvement in efficacy or toxicity compared to standard regimens with paclitaxel alone[2][4][6]. The development of this combination for these indications has not progressed due to lack of added benefit.
