Overview

A combination therapy of **inlexisertib** (DCC-3116), an investigational selective autophagy inhibitor, and **sotorasib** (AMG 510, marketed as Lumakras), an FDA-approved selective KRAS G12C inhibitor. This combination is being studied for its safety and efficacy in patients with non-small cell lung cancer (NSCLC) harboring a KRAS G12C mutation. Inlexisertib targets and inhibits the ULK1/2 kinases, suppressing autophagy, which may enhance tumor cell death. Sotorasib binds irreversibly to KRAS G12C, inhibiting its oncogenic signaling. The combination aims to overcome acquired resistance to sotorasib by targeting parallel resistance pathways[3][5].
