INO-3106 + INO-9012 is an investigational combination immunotherapy developed by Inovio for the treatment of HPV type 6-associated diseases, particularly recurrent respiratory papillomatosis (RRP) and other aerodigestive tract precancers and cancers. INO-3106 is a DNA plasmid-based immunotherapy encoding the E6 and E7 proteins of human papillomavirus type 6 (HPV 6), designed to elicit a robust T cell-mediated immune response against HPV 6-infected cells. INO-9012 is a DNA plasmid encoding interleukin-12 (IL-12), which acts as an immune activator to enhance cytotoxic T lymphocyte responses when co-administered with antigen-targeting vaccines like INO-3106. Both components are delivered intramuscularly using electroporation via Inovio’s CELLECTRA device, which facilitates cellular uptake of the DNA plasmids. The combination has demonstrated safety, tolerability, immunogenicity, and preliminary clinical efficacy in early-phase trials for patients with HPV 6-positive RRP and related tumors[1][2][5][7].
