Overview

INO-5401 + Libtayo is a combination immunotherapy regimen consisting of INO-5401, a synthetic DNA medicine, and cemiplimab (Libtayo), a programmed death receptor-1 (PD-1) blocking monoclonal antibody. INO-5401 is designed to activate the immune system against multiple tumor-associated antigens, including human telomerase reverse transcriptase (hTERT), Wilms tumor gene 1 (WT1), and prostate-specific membrane antigen (PSMA). Cemiplimab acts as a checkpoint inhibitor to prevent the PD-1/PD-L1 interaction, thereby enhancing T-cell mediated anti-tumor responses. This combination is primarily being investigated for the treatment of newly diagnosed glioblastoma (GBM) to improve survival outcomes by priming and sustaining a robust immune response against the tumor. The regimen often includes INO-9012, a DNA plasmid encoding interleukin-12 (IL-12), which serves as an immune booster.
