Overview

This is an investigational combination immunotherapy regimen evaluated for advanced cancers, particularly *urothelial carcinoma*. **INO-5401** is a DNA vaccine consisting of synthetic plasmids encoding three tumor antigens: Wilms tumor gene-1 (WT-1), prostate specific membrane antigen (PSMA), and human telomerase reverse transcriptase (hTERT), designed to generate **antigen-specific T cell responses**. **INO-9012** is a DNA plasmid encoding human interleukin-12 (IL-12) to enhance immune activation. **Atezolizumab** is a fully humanized IgG1 monoclonal antibody that inhibits **programmed cell death ligand 1 (PD-L1)**, resulting in immune checkpoint blockade and restoration of antitumor T cell responses. The combination is delivered by intramuscular injection followed by electroporation (INO-5401, INO-9012) and intravenous atezolizumab. The regimen is in clinical development for patients with advanced, unresectable, or metastatic urothelial carcinoma, including those who progressed after or are ineligible for platinum-based therapy[1].
