inotuzumab ozogamicin + standard maintenance therapy

This is a combination regimen consisting of inotuzumab ozogamicin, an antibody-drug conjugate targeting CD22, plus standard maintenance therapy for acute lymphoblastic leukemia (ALL). Inotuzumab ozogamicin is composed of a humanized IgG4 monoclonal antibody directed against CD22 and covalently linked to the cytotoxic agent N-acetyl-gamma-calicheamicin. The antibody binds to CD22-expressing B-cell precursor leukemic cells, leading to internalization and intracellular release of calicheamicin, which induces double-stranded DNA breaks and apoptosis. Standard maintenance therapy typically refers to prolonged low-intensity chemotherapy regimens such as POMP (purinethol [6-mercaptopurine], oncovin [vincristine sulfate], methotrexate, prednisone), used after induction/consolidation phases in ALL treatment[6][8]. This combination has been studied particularly in older adults with newly diagnosed or relapsed/refractory B-cell precursor ALL.
