Overview

inotuzumab ozogamicin + temsirolimus is a combination therapy investigated for the treatment of relapsed or refractory CD22-positive B-cell non-Hodgkin lymphoma. The combination consists of inotuzumab ozogamicin, a CD22-directed antibody-drug conjugate (ADC), and temsirolimus, a small molecule inhibitor of the mammalian target of rapamycin (mTOR). Inotuzumab ozogamicin (Besponsa) targets CD22-expressing B-cells and delivers a potent cytotoxic payload, calicheamicin, which induces double-strand DNA breaks and subsequent apoptosis. Temsirolimus (Torisel) inhibits the mTOR kinase, a central regulator of cell growth, proliferation, and survival that is often dysregulated in lymphomas. By combining targeted cytotoxicity with pathway inhibition, the regimen aims to overcome resistance and improve efficacy in patients with aggressive B-cell malignancies.
