IGF1-uIONP/SN38 is an experimental nanoparticle-based drug delivery system designed for the targeted treatment of pancreatic cancer. It consists of ultrafine iron oxide nanoparticles (uIONP) conjugated with insulin-like growth factor 1 (IGF1) as a targeting ligand and loaded with SN-38, the potent active metabolite of the chemotherapy drug irinotecan. The platform leverages the overexpression of the insulin-like growth factor 1 receptor (IGF1R) in pancreatic cancer cells for active targeting. Upon receptor-mediated internalization, the system utilizes a pH-responsive mechanism to release SN-38 in the acidic environment of the tumor interstitium and lysosomes. SN-38 acts as a topoisomerase I inhibitor, inducing DNA damage and apoptosis in cancer cells. This theranostic platform aims to overcome the delivery barriers posed by the dense stroma of pancreatic tumors while providing potential for imaging applications through the iron oxide core.
