INT230-6 + pembrolizumab is an experimental combination therapy for advanced solid tumors, currently in clinical development. INT230-6 consists of two cytotoxic agents, cisplatin and vinblastine, co-formulated with an amphiphilic cell penetration enhancer for direct intratumoral injection, designed to facilitate drug diffusion into tumor cells and maximize drug retention within tumors. Pembrolizumab is a monoclonal antibody targeting the programmed death receptor-1 (PD-1), employed as an immunotherapy checkpoint inhibitor. The combination aims to induce local tumor necrosis with subsequent immune activation, leading to both local and systemic anti-tumor effects. Early clinical results suggest that INT230-6 promotes a local cytotoxic response and neoantigen release, augmenting immune cell infiltration into tumors (including CD4 and CD8 T-cells), and pembrolizumab potentiates the systemic immune response by checkpoint inhibition[1][2][3][4][5].
