Intensified TB treatment (DATURA)

The Intensified TB treatment (DATURA) is a multi-drug therapeutic regimen being evaluated in the Phase III DATURA clinical trial (ANRS 12424). It is specifically designed for HIV-infected adults and adolescents hospitalized with tuberculosis and severe immunosuppression (CD4 count ≤ 100 cells/μL). The regimen consists of an initial 8-week phase using increased doses of rifampicin (35±5 mg/kg daily) and isoniazid (10±2 mg/kg daily), combined with standard doses of pyrazinamide and ethambutol. Additionally, the regimen includes adjunctive corticosteroid therapy with prednisone for 6 weeks and a 3-day course of albendazole. This intensified approach aims to improve survival rates in this high-risk population by providing more potent initial antimicrobial activity and managing the inflammatory response associated with severe TB-HIV co-infection.
