Overview

This combination therapy is an experimental immunotherapeutic regimen developed by the Department of Infectious Diseases at the Third Affiliated Hospital of Sun Yat-sen University for the treatment of advanced hepatocellular carcinoma (HCC). The protocol involves the administration of autologous dendritic cells (DCs) that have been pulsed with HepG2 cell lysate to present a broad spectrum of tumor-associated antigens to the patient's immune system. This is combined with recombinant interferon-alpha (IFN-α), which promotes DC maturation and enhances the cytotoxic activity of T cells and natural killer cells. Additionally, low-dose (metronomic) cyclophosphamide is administered to selectively deplete CD4+CD25+ regulatory T cells (Tregs), thereby reducing immune suppression within the tumor microenvironment and potentiating the vaccine-induced anti-tumor response.
