This combination therapy regimen consists of the immunomodulatory cytokine interferon alfa-2a added to the standard ABVD chemotherapy protocol, which includes doxorubicin (Adriamycin), bleomycin, vinblastine (Velban), and dacarbazine. Investigated by the M.D. Anderson Cancer Center in a Phase II clinical trial (NCT01404936), the regimen was designed for patients with relapsed or advanced untreated Hodgkin lymphoma. The rationale for adding interferon alfa-2a is to leverage its immunomodulatory and antiproliferative properties to enhance the anti-tumor immune response against Hodgkin's disease. Interferon alfa-2a is a recombinant protein that binds to type I interferon receptors (IFNAR1 and IFNAR2), while the ABVD components provide multi-targeted cytotoxic effects through DNA intercalation, DNA alkylation, DNA cleavage, and microtubule inhibition.
