Overview

Interferon alfa-2a + retinoic acid is a combination therapy evaluated by the National Cancer Institute (NCI) in a Phase 2 pilot study for the treatment of lymphoproliferative disorders in children with congenital or acquired immunodeficiency syndromes, including HIV. Interferon alfa-2a is a recombinant type I interferon that binds to the IFNAR1/IFNAR2 receptor complex, triggering Janus kinase-signal transducer and activator of transcription (JAK-STAT) signaling to exert immunomodulatory, antiproliferative, and antiviral effects. Retinoic acid (initially 13-cis-retinoic acid/isotretinoin, later all-trans-retinoic acid/tretinoin) acts as a differentiating agent by binding to retinoic acid receptors (RARs), promoting the maturation of lymphoid cells and inhibiting their uncontrolled proliferation. This combination was specifically targeted at conditions like diffuse infiltrative lymphocytosis syndrome (DILS) and polyclonal B-cell lymphoproliferative disorders in immunocompromised patients.
