Overview

Interferon alfacon-1 + ribavirin is a combination therapy used primarily for the treatment of chronic hepatitis C in adults with compensated liver disease. Interferon alfacon-1 is a recombinant type I interferon that binds to interferon alpha/beta receptor 1 (IFNAR1) and interferon alpha/beta receptor 2 (IFNAR2), activating Janus kinase 1 (JAK1) and tyrosine kinase 2 (Tyk2) and signal transducers and activators of transcription (STATs), leading to the expression of immunomodulatory and antiviral proteins. Ribavirin is a nucleoside analog with multiple mechanisms, including inhibition of inosine monophosphate dehydrogenase (IMPDH) resulting in depletion of GTP pools, direct inhibition of viral replication, induction of mutagenesis in viral genomes, and immunomodulation by shifting immune response toward Th1 phenotype. The combination has shown improved sustained virologic response rates compared to other interferons plus ribavirin regimens, especially in patients with genotype 1 hepatitis C virus infection and high viral load[2][3][4][5][6].
