Interferon alpha-2a + donepezil is an investigational combination therapy studied for the treatment of mild to moderate Alzheimer's disease. The regimen consists of human recombinant interferon alpha-2a (hrIFN-a), a cytokine typically used for its antiviral and antineoplastic properties, administered orally (ingested) alongside the standard-of-care acetylcholinesterase inhibitor donepezil (Aricept). The National Center for Research Resources (NCRR) sponsored a Phase I/II clinical trial (NCT00031018) to evaluate whether low-dose ingested interferon alpha could modulate neuroinflammation by decreasing acute-phase reactants and pro-inflammatory cytokines like interleukin-6 (IL-6), thereby slowing cognitive decline. While interferon alpha-2a is well-established in other therapeutic areas, its development for Alzheimer's disease has not progressed beyond early-phase clinical investigation.
