Overview

This is a triple combination therapy consisting of interferon alpha-2b, ribavirin, and amantadine. Interferon alpha-2b is a recombinant cytokine with antiviral and immunomodulatory properties; it stimulates the immune response against viral infections. Ribavirin is a nucleoside analog that inhibits viral RNA synthesis and replication. Amantadine is an antiviral agent that blocks the uncoating of certain viruses by inhibiting the M2 protein ion channel, though its mechanism against hepatitis C virus (HCV) remains unclear. This combination has been studied primarily for retreatment in patients with chronic hepatitis C who have failed previous therapies with interferon-based regimens[1][2][4]. Clinical studies indicate that while this triple therapy can induce biochemical and virological responses in some patients, sustained virological response rates are generally low (10–19%) in interferon-resistant or non-responder populations[1][2][4][5].
