This drug combination involves the sequential or comparative administration of interferon-beta and natural human leukocyte interferon-alpha (specifically the brand Multiferon). It was investigated in the REPAIR (Response to IFN-alpha in IFN-beta Neutralising Antibody Positive Multiple Sclerosis Patients) Phase 2 clinical trial led by Melinda Magyari at the Danish Multiple Sclerosis Center. The treatment strategy targets patients with relapsing-remitting multiple sclerosis (RRMS) who have developed neutralizing antibodies (NAbs) against interferon-beta, which typically renders that therapy ineffective. Both interferon-alpha and interferon-beta are Type I interferons that signal through the same heterodimeric receptor complex (IFNAR1/IFNAR2). The combination or switch to interferon-alpha is intended to bypass the specific neutralizing antibodies directed at interferon-beta, thereby restoring the induction of interferon-stimulated genes like MX1 and maintaining therapeutic efficacy.
