**interferon beta + ribavirin** is a combination antiviral therapy primarily investigated for the treatment of chronic hepatitis C and COVID-19. Interferon beta is a cytokine in the interferon family that modulates immune responses and has antiviral properties, primarily by upregulating proteins such as PKR, which inhibit viral replication. Ribavirin is a nucleoside analog that inhibits viral RNA synthesis and induces phosphorylation of proteins important in shutting down viral translation, also depleting guanosine pools via inhibition of IMPDH. The two drugs act synergistically: interferon beta induces PKR activation leading to eIF2α phosphorylation and inhibition of ribosome loading, while ribavirin inhibits IMPDH, lowering GTP pools, both contributing to inhibition of viral RNA replication and translation—their combination shows enhanced efficacy over either agent alone in both hepatitis C and emerging viral infections like COVID-19[3][4][5][1].
