This is a combination therapy of three drugs: **interferon beta-1a** (a recombinant protein immunomodulator approved for relapsing forms of multiple sclerosis), **methotrexate** (a folate antimetabolite with immunosuppressive and anti-inflammatory properties), and **methylprednisolone** (a synthetic glucocorticoid corticosteroid with potent anti-inflammatory and immunosuppressive effects). The combination has been studied in relapsing-remitting multiple sclerosis (RRMS) patients with breakthrough disease activity on interferon beta-1a monotherapy. The rationale is to leverage non-redundant immunomodulatory mechanisms to enhance disease suppression. Clinical studies, including the Avonex Combination Trial (ACT), indicate that the triple combination is generally safe and well tolerated but did not demonstrate significant clinical benefit over monotherapy, with only modest trends in reduced relapse rates and MRI lesions. This regimen is not an FDA-approved fixed-dose combination; it is used off-label in specialized clinical protocols for difficult-to-control MS cases[1][3][5][7].
