Interleukin-10 + 2-Trikine (IL-10/2-Trikine) is an engineered biologic immunotherapeutic developed by researchers at Stanford University. It belongs to a novel class of molecules called 'Trikines,' which are designed to compel the cis formation of non-natural three-chain cytokine receptor complexes at the cell surface to induce customized STAT transcriptional signaling programs. This specific IL-10-based Trikine consists of interleukin-10 linked to a binder that recruits the interleukin-2 receptor beta chain (IL-2Rβ). By assembling a trimeric complex of IL-10R1, IL-10R2, and IL-2Rβ, the molecule 'rewires' intracellular signaling to activate both the natural pSTAT3 pathway of IL-10 and a proinflammatory pSTAT5 signature. In preclinical models, IL-10/2-Trikine has demonstrated the ability to induce immune cell infiltration into poorly immunogenic tumors and has shown significant efficacy in small cell lung cancer and pancreatic cancer models, particularly when combined with PD-1 checkpoint blockade.
