Overview

This is a combination immunotherapy regimen consisting of three agents: - Interleukin-2 (IL-2): a cytokine therapy that stimulates the proliferation and activation of T-cells, enhancing the immune response against cancer cells. - Ipilimumab: a monoclonal antibody checkpoint inhibitor targeting cytotoxic T lymphocyte-associated protein 4 (CTLA-4), thereby promoting T-cell activation and proliferation. - Nivolumab: a monoclonal antibody checkpoint inhibitor targeting programmed cell death protein 1 (PD-1), which prevents tumor-induced inhibition of T-cell activity. The combination aims to synergistically enhance anti-tumor immunity by both directly stimulating immune effector cells (via IL-2) and releasing inhibitory checkpoints on T-cells (via ipilimumab and nivolumab). This approach is under investigation for advanced cancers, particularly melanoma, where dual checkpoint blockade with ipilimumab plus nivolumab has demonstrated improved outcomes compared to monotherapy. The addition of interleukin-2 may further augment immune-mediated tumor destruction.
