A combination therapy consisting of low-dose interleukin-2 (IL-2) and methotrexate (MTX) used primarily for the treatment of rheumatoid arthritis (RA). This combination has shown superior efficacy compared to MTX alone in clinical outcomes including CDAI, SDAI, and ACR response. Low-dose IL-2 works by increasing regulatory T cells (Tregs), which helps maintain immune balance, while MTX has anti-inflammatory effects related to interruption of adenosine and other inflammatory pathways. The combination therapy has demonstrated significant reductions in inflammatory cytokines, including IL-17A, IFN-γ, TNF-α, and IL-12 levels.
