Overview

This is a combination immunotherapy consisting of two recombinant cytokines, interleukin-2 (IL-2) and tumor necrosis factor alpha (TNF-α). Both are biologic agents with distinct but complementary mechanisms in modulating the immune response against cancer. IL-2 stimulates the proliferation and activation of T lymphocytes and natural killer (NK) cells, enhancing cell-mediated cytotoxicity. TNF-α is a pro-inflammatory cytokine that can induce direct tumor cell apoptosis, disrupt tumor vasculature, and further activate immune effector functions. Preclinical studies have shown that this combination has synergistic antitumor effects in various cancer models, leading to enhanced regression compared to either agent alone. Clinical trials have evaluated this combination primarily for advanced malignancies such as non-small cell lung cancer and melanoma, demonstrating measurable tumor regression in some patients with acceptable toxicity at low doses[4][6][7]. The most common side effects include fever, local skin reactions at injection sites, pancytopenia, malaise, and dose-limiting thrombocytopenia; these are generally reversible upon cessation of therapy[6]. This drug is administered via intravenous infusion (IL-2) combined with intramuscular or intravenous TNF-α.
