Overview

This is a combination therapy of interleukin-2 (IL-2), a recombinant cytokine that stimulates the proliferation and activation of T lymphocytes, and zoledronate (zoledronic acid), a potent aminobisphosphonate. The combination is designed to augment the expansion and activation of Vγ9Vδ2 T cells—a subset of γδ T cells with antitumor activity—by leveraging the synergistic effects of both agents. Zoledronate induces accumulation of phosphoantigens in tumor cells, which activates Vγ9Vδ2 T cells; IL-2 further promotes their proliferation and cytotoxic function. This regimen has been investigated primarily as maintenance therapy after autologous stem cell transplantation in multiple myeloma, as well as for recurrent/metastatic renal cell carcinoma and other malignancies[1][4][3]. The approach aims to enhance immune-mediated tumor control by expanding effector γδ T-cell populations.
