Overview

This is a combination therapy consisting of an interleukin-6 (IL-6) antagonist and a corticosteroid. IL-6 antagonists, such as tocilizumab and sarilumab, are monoclonal antibodies that block the activity of the pro-inflammatory cytokine IL-6 by inhibiting its receptor or ligand. Corticosteroids are synthetic analogs of endogenous glucocorticoids that suppress inflammation and modulate immune responses through multiple mechanisms, including inhibition of cytokine production. The combination is primarily used in severe or critical COVID-19 to reduce hyperinflammation ("cytokine storm") and improve survival outcomes. Clinical evidence shows that this combination reduces mortality more effectively than either agent alone in hospitalized patients with severe COVID-19[2][4][5]. The approach may also be relevant for other diseases characterized by excessive inflammation.
